Illuminating Hidden Cellular Pathways During Differentiation of Dendritic Cells

 

The adaptive immune system has evolved sophisticated ways to detect and eliminate malignantly or virally transformed cells. Malfunctioning can lead to chronic infection and autoimmunity. We will explore molecular machines in cellular compartmentalization as well as MHC I-chaperone and ER-quality control complexes in adaptive immunity and also in terms of viral immune evasion. The key steps in these pathways and the mechanisms of molecular machines involved are largely unknown. This emerging research area will be addressed by integrative approaches comprising single-cell biochemistry, CRISPR/Cas9 technology (genomic trapping of native complexes), antibody engineering, chemical biology, super-resolution microscopy, and single-molecule spectroscopy.
